Promoter Group
Transformation of Ligation
Thaw TOP10 supercompetent cells on ice.
Add 5 ul of Fe/mRFP ligation. Tap tube to mix.
Incubate on ice 30'.
Heat shock cells @ 42C for 45.
Incubate on ice 2'.
Transfer cells + DNA to 1.5 ml tube w/ 900 ul 2XYT.
Incubate 1h @ 37C w/ shaking.
Plate 100 ul cells undiluted and 100 ul cells concentrated.
