Today we are researching PIF3 degredation as it might occur in Ecoli
Writing the new fundraising letters
Finishing construction of our biobricks
Colony PCR,Gel extraction, and streaking positive colonies and setting up O/N cultures of last years project.
- Ran the thiolase sequencing that I did last week up to the MBSU. We should get the results back tomorrow.
- Finished up the BioBricks we've worked on so far: PIF3 and the Phytochromobilin/Heme Oxygenase Operon. The HIS-tags have been added to all three genes and they have been optimized for expression in E.coli (strain K12, specificly, though we are going to be using XL10. The optimization site I mentioned in the May 16th notes did not have the XL10 strain listed so I went with K12. I don't anticipate this being a problem). The
Phytochromobilin/Heme Oxygenase operon looks like this:
The black squares at either end are the standard BioBrick suffix and prefix. The blue represents a terminator sequence (part BBa_B1001) and the red diamonds mark where we put in the RBS (part BBa_B0034).
- Did some editing to our team's wiki front page. The image now links directly to the UofA website. Jason made a new Nav Bar and I was able to get the images to link to their corresponding pages. The nav bar on the Notebook and Project pages are not aligned in the center despite the pages having the center alignment tag. If anyone knows how to fix this, then be my guest. I omitted the Nav Bar from the Modelling and Parts pages since it didnt seem to be working correctly on those pages, and we have yet to use them anyway. It can be fixed later.

Lab Tip of The Day
When inserting a DVD into a MAC computer, please ensure that you have access to said computer, or else your DVD will become lost. Please remember to carry a 2008 Official iGEM Pointed DVD Removal Device (a.k.a a stick) with you at all times to aid in DVD removal in the case of catastrophic DVD loss.
