  • Minipreps M198 to M209:
    • M198 to M200: L58 = self-ligation of P91 (SOB2-SU1). M199 and M200 were missed out by accident.
    • M201 to M203: L59 = self-ligation of P92 (SOB2-rbs-SU1).
    • M204 to M206: L60 = self-ligation of P93 (SOB2-ISO2).
    • M207 to M209: L61 = self-ligation of P94 (SOB2-rbs-ISO2). (Yan)
    • M198, M201 to 209 subsequently digested with EcoRI and PstI. (AM)
  • Re-ran M182~187 (pSB1A2+cenA) on Gel 72 and confirmed failure of pSB1A2+cenA ligation. (AM)
  • Ran P95 and 'cex mut' (both cex mutagenic PCR) on Gel 73. Both failed. (AM)
  • Ligations L63 to L64:
    • L63: P90 (CHU2268 bglX) to Edinbrick 1.
    • L64: P65 (cenA) to Edinbrick 1. (AM)
  • Submitted M161 and M162 (pSB1A2+dxs+lims+appY) for sequencing using reverse primer only. (AM)
