Things we did today

Wetlab work

Plasmid prep

EtBr stained 0.8% agarose gel ran at 95V for 1 hour. Ten microliters of plasmid were loaded into each well.
Grace
  • Used Qiaprep Spin Miniprep Kit to plasmid prep BB1 (10/22 and 10/25 inoculations)
  • Ran on gel to verify plasmid
  • ~2ng/μl
  • The prep kit yields really clean preps in ~1 hour. :D!

Plasmid prep

EtBr stained 0.8% agarose gel ran at 95V for 1 hour. 5 microliters of plasmid were loaded into wells 3 and 4 while ligation products were loaded into lanes 5 and 6.
Margaret

Construction of BB-pRL1383a/K125820

Krystle

  • Restriction digest BB-pRL1383a with EcoRI and PstI
  • Dephosphorelated restriction product with SAP for 1 hour
  • Ligated cut/dephosphorelated product with gel purified K125820 using T4 ligase
