To Do:

  • check to see if Ni-Columns have arrived
  • Finish westerns

Today:

  • Checked the transformants Anthony did on the weekend. The Purple Russian had no growth at all :( Tryp had what appeared to be satellite colonies; we did Colony PCR to check to see if they have an insert. Blue Ox had pleanty of colonies, so hopefully we have gotten that promotor out and have a complete BioBrick now. EDIT: I ran the colony PCR out on a gel and imaged it - there definately was a band, but it was somewhere around 900bp in length; the Tryp biobrick should be around 1400bp. Im not sure what it is supposed to be. Its too big to be GFP (and the colonies dont glow green). I guess we could sequence it tomorrow and try to figue out what it is. -Cwk 21:53, 9 June 2008 (UTC)
  • Winnie did sequencing on the Blue Ox colonies just to make sure that they are what we think they are.
  • The result of Anthony's experiment with transforming Purple Russian plates in tinfoil was inconclusive. There was no growth on the plates, but since there was no growth on the plates that were in the incubator as well, we cannot conclude that the lack of colonies was due to the tinfoil technique itself. We should try it again.
