- P96: Repeat of MABEL PCR of cex with glycerol, M150 as template. Denaturing time 1 min, extension time 85 seconds. Run on Gel 74 - results indicate the presence of cex and pSB1A2. (YAN)
- Yesterday's transformations failed. Hence, repeated transformations with:
- L62 (dxs+crtE into crtBI) --> Plates 188~189.
- L63 (pSB1A2-bglX) --> Plates 190~191.
- L64 (pSB1A2-cenA) --> Plates 192~193.
- Edinbrick 1 as control --> Plate 194 (900µl). (AM)
- Sequencing results reveal that M162 (putative pSB1A2-dxs+lims+appY) does indeed include appY. Set up overnight culture of cells transformed with pSB1A2+dxs+lims+appY (from Plate 155, Streak 3) for maxiprep tomorrow, expected to be analogous to M162. (AM)
