Today we (Dave, Lisa and Tin)isolated our pUC-EUY and pAC plasmids from broth cultures made from the transformed E. coli of June 5th. We used a commercial prep kit to do this, and made a total of six preps, three from the pAC and three from the pUC-EUY.

Upon isolation, we then ran a restriction digest using HindIII to confirm that we had the plasmids we were looking for, as pUC-EUY has no HindIII sites, and pAC has two, resulting in a single band for pUC-EUY and a double band for pAC.

We would have had clearer results if we'd run our gels a little longer, but there was enough evidence present to satisfy us that we had the right plasmids.
