1. Ligation Transformations SUCCESSFUL: 3 out of 4 plates had growth.
2. Pick Colonies from Transformation Plates: Place picked colonies in 2mL LB cultures. Allow 2 hours to incubate and grow @ 37C with shaking @ 225rpm.
3. Plasmid Prep: Plasmid prep the 2mL cultures of ligation products. This is done to extract DNA.
4. Plasmid Prep Promoters and Terminators
