Digested SOS plasmids:
- Mixture:
- 40uL DNA
- 5uL NEB Buffer 2 (10x)
- 13uL ddH2O
- 0.5uL BSA
- 1uL EcoRI
- 1uL SpeI
- Mixed, stored at 37C for 1 hour
- Placed in -20C to halt reaction
We'll run it through a gel and purify the SOS and LacZ parts soon. Then ligation, transformation, testing!
