- Overnight culture of p1010 A2 (LB+Amp) and p1010 K3 (LB+Kan), i.e. 200µl of little overnight culture in 100 ml medium
- Overnight culture of Top10 cells from Invitrogen (cultivated on a plate) to make them competent tomorrow
- PCR of VP16 (3 different templates) Tup1,LacI,CI,TetR @ 48,50,52,53 °C -> none worked
