  2. re-digest pPIC6 #2
  3. prepare and run 1% agarose gel
  4. make now low salt plates
  5. purify gel cut-out
  6. make ligation mix using T4, Lip, and pPIC6
  7. perform transformation with ligation mix
  8. spread onto plates for overnight growth
