  • Gel purification of 1) conc. double digests of BisdA and BisdB with both (NgoMIV+Age1) and (XbaI+PstI); 2)conc. digest of vector containing the TetR binding site and the RBS with NgoMIV (digest was treated with Antartic Phosphatase); 3) conc. digest of J61003 with (SpeI+PstI) (digest was treated with Antartic Phosphatase).
  • Minipreped TetR O/N's set up on Tuesday night. It is in MininPrep Box 3.
  • Digested B0034 with Spe/Pst; TetR with XbaI/Pst. Digested the biobrick Lac1-ERE with Spe/Pst. Running them on 1% gel.
    • Gel purified the digestions.
  • Ligated TetR with RBS (B0034).
  • Digested pTet (R0400) with Xba/Pst.
