  1. Transformation of E. coli TOP10 carrying pZC320 with:
  2. Plating transformants on 'TAXI' (LB + Tetracycline + IPTG + Ampicillin 30 μg/mL + X-Gal).
  3. Inoculation of transformants to liquid LB + Tetracycline + Ampicillin 30 μg/mL +/- 0.1% Ara.
