Six ligations were performed, three with new T4 ligase and three with older T4 ligase: -35 Pro in pSB1A3, 5'F/SD R in pSB1A3, and phaB in pSB1A3.
Cryostocks were made of 5'F/ATG R, 2x 5'F/TC R, and -35F/TC R
