  • Cut A77, A79, A80 (E,S) to assemble with terminator
    • Rationale is that maybe having the promoter in front of the coding region with no terminator is killing the cells
  • Gelled A82(3) (E,P) again, no band for cut-out, leads us to believe vector is not correct
  • Running more of the RBS+FP (X,P) DNA through low melt gel to ligate straight from gel instead of from gel-extracted DNA
