Overnight Results
- pSB1A10 screening plasmid grew just fine
- Assembly 1 (BP1 + Term) grew more colonies
- pSB1AC3 and pSB1AK3 both grew a small number of colonies
Goals
- Transform overnight ligations
- Note: Considering the other ligations didn't work and the restriction digest might be the problem we don't expect much
- Make small (10ml) broth of plates with colonies
- pSB1AC3, pSB1AK3
- Repeat assemblies
- Doing digestion according to the [http://openwetware.org/wiki/Knight:Restriction_Digest Knight protocol] now
