Goals
- small broth of plated terminator
- large broth of E0240(RFP) and I715039(GFP)
- mini prepe PRE RFP and RET GFP for another ligation
- redigest 6 parts for the three ligations
- purify digestions
- run NuSieve gels to verify parts
- three runs of 3A assembly
- RFP+GFP
- Promoter+RBS
- BP1+terminator
Notes
- previous trials of 3A assembly failed
- digestion should have been fine so either ligation or purification steps went wrong
- iGEM agreed to send us CRUCIAL non functioning part from last year's library
- great success!!
