lab work (SD):
Digested K102002.1-.6 and freshly annealed TA0 with BamHI + NcoI. Incubated at 37 C for 2 hrs. Ran K102002.1-.6 on 2% agarose gel at 110V for 50 min. Purified TA0 using QIAquick nucleotide removal protocol. Quantified DNA for K102002 and TA0. Ligated TA0 into K102002.1-.6 and incubated at 16 C for 30 min.
