  • Miniprepped 15mL culture of 3:1 ligation.
  • Made a glycerol stock.
  • Restriction digest done of the ligation to make sure the ligation was successful.
  • Restriction Digest:
  1. 18 ul Water
  2. 25 ul DNA (82.5 ng/ul)
  3. 1 ul HindIII
  4. 1 ul EcoRI
  • Ran a gel of the digest with uncut DNA.
  • Results:
  • One band at approximately 5 kb
  • One band at 3-4 kb
  • A faint band possibly at <100 bp
