- Miniprepped 15mL culture of 3:1 ligation.
- Made a glycerol stock.
- Restriction digest done of the ligation to make sure the ligation was successful.
- Restriction Digest:
- 18 ul Water
- 25 ul DNA (82.5 ng/ul)
- 1 ul HindIII
- 1 ul EcoRI
- Ran a gel of the digest with uncut DNA.
- Results:
- One band at approximately 5 kb
- One band at 3-4 kb
- A faint band possibly at <100 bp
