Product Transport in E. Coli (E. Coli Filter)

  • Goal: Make E. coli absorb some product.
  • Questions:
    • What products do we want E. coli to absorb?
    • How should E. coli absorb the products?
  • Notes:
    • Someone should research [http://en.wikipedia.org/wiki/Bioremediation bioremediation] to determine what products cause problems.
    • We should understand transport across the cell envelope of [http://en.wikipedia.org/wiki/Gram-negative Gram-negative bacteria].
    • E. coli do transport iron across their cell envelope. A great review is [http://www.sciencemag.org/cgi/reprint/295/5560/1658.pdf Close Before Opening].
    • A starting point for other metals is [http://www.sciencedirect.com/science?_ob=ArticleURL&_udi=B6T1W-4JTJ9T5-1&_user=616147&_rdoc=1&_fmt=&_orig=search&_sort=d&view=c&_acct=C000032324&_version=1&_urlVersion=0&_userid=616147&md5=a755ffdf33ea1c8d5f687b7385e3f250 Structural Analysis and Classification of Native Proteins from E. Coli Commonly Co-Purified by Immobilised Metal Affinity Chromatography].

Chemotaxis Control (E. Coli Filter and E. Coli Rugby)

  • Goal: Make E. coli follow/flee some chemical attractant/repellent.
  • Questions:
    • How does the chemotactic system work?
  • Notes:
    • Read [http://pubs.acs.org/cgi-bin/sample.cgi/jacsat/2007/129/i21/pdf/ja0692480.pdf Guiding Bacteria with Small Molecules and RNA].

Mechanical Stress Detector

  • Goal: Make E. coli respond to mechanical stress.
  • Questions:
    • How does [http://en.wikipedia.org/wiki/Mechanotransduction mechanotransduction] work in E. coli?
  • Notes:
    • Sound waves are mechanical stress, so this is an acoustic sensor. We would have to consider some frequency range.
    • Read [http://www.childrenshospital.org/research/ingber/PDF/2002/STKE-Alenghat.pdf Mechanotransduction: All Signals Point to Cytoskeleton, Matrix, and Integrins] and [http://www.proteinscience.org/cgi/content/abstract/8/10/1915 Structure and Function of the Bacterial Mechanosensitive Channel of Large Conductance].

Lipopolysaccharide Detector

  • Goal: Make E. coli detect lipopolysaccharide (LPS).
  • Notes:
    • LPS leads to inflammation in higher organisms.

Light Sensor (E. Coli Sunscreen)

  • Goal: Make E. coli respond to varying intensities of light.
