- Amplification of LacI-GFP ([http://partsregistry.org/wiki/index.php?title=Part:BBa_J04430 BBa_J04430]) and Terminator ([http://partsregistry.org/wiki/index.php?title=Part:BBa_B0015 BBa_B0015]) parts.
- Overnight incubation at 37 degrees C of these 3 agar plates:
  1. LB AmpR spread with 250ul cells with LacI-GFP plasmid.
  2. LB AmpR spread with 125ul cells with Terminator plasmid.
  3. LB KanR spread with 125ul cells with Terminator plasmid.
