  • PCR confirms the identy of the Gal4O, LacIO, LexAO, cIO, tetO BioBricks (Which are in the pSB1A2 backbone and in the Silver-standard)
  • Miniprep of the overnight cultures
  • Digestion with SpeI and PstI of the LacIO, LexAO, cIO and the tetO vectors -> back vectors
  • Ethanolic precipitation to get ride of the small fragments
