  • M144-M147: Minipreps of cenA(1,2,3,4). M148-M151: Minipreps of cex(1,2,3,4). M152-M155: Munipreps of dxs-crtE (1,2,3,4). Another mistake made, I used methanol(920ul) instead of ethanol for M144-M147.(OG)
  • Minipreps 144-155 were digested with EcoRI/PstI. This need running on a gel tomorrow. (AH)
  • X11: Maxiprep of Zea mays isoamylase 1 (isa1+pZntr22). X12: Maxiprep of Zea mays isoamylase 2 (isa2+pZntr22). (HX)
  • Combined biobricks:

L42: dxs+lims (M137) to appY (M82). L43: crtB+crtI (M109) to appY (M82). L44: pstA (M130) to lacB. (YAN)

  • Blunt-ended ligations L45~L47 of purified P71A (glgC mut 1+2), P71 and P72 (both glgC mut 1+2+3) to SOB2 ("son of BABEL 2"). (AM)
  • Prepared competent cells (labelled 'iGEM JM109 18/8 1-35') 1-24 = 100 microlitres each. 26-35 = 200 microlitres each. Stored at -80C. (AH, AM)
