  • 10 mL cultures of Ry Young's DNA were made (Swt, S105, and S105 A25G).
  • Glycerol Stocks made.
  • Cultures were then miniprepped:
  1. pSWT 1: 107.2 ng/ul
  2. pS105 1: 136.5 ng/ul
  3. pS105 2: 161.2 ng/ul
  4. pS105 A25G 1: 110.2 ng/ul
  5. pS105 A25G 2: 110.4 ng/ul
  • PCR will be performed on minipreps, followed by ligation and transformation.
