Goals

  • small broth of plated terminator
  • large broth of E0240(RFP) and I715039(GFP)
  • mini prepe PRE RFP and RET GFP for another ligation
  • redigest 6 parts for the three ligations
  • purify digestions
  • run gels to verify parts
  • three runs of 3A assembly
    • RFP+GFP
    • Promoter+RBS
    • BP1+terminator


Notes

  • previous trials of 3A assembly failed
    • digestion should have been fine so either ligation or purification steps went wrong
  • iGEM agreed to send us CRUCIAL non functioning part from last years library
    • great success!!
