Things we did today
Wetlab work
PCR
- Margaret
- Ran 10 μl PCRs for oriV, omega, rep, and mob
Purified I14032
- Grace
- Plate had incubated ~60 hrs. 48 colonies observed (satellites? kanamycin broke down?). Restreaked on fresh LB+kan50 plate.
Grew transformed E. coli for cryostocking
- Grace
- Inoculated 5 ml LB+amp100 with one colony of each of last week's Biobrick transformed E. coli
- Incubated overnight at 37C with shaking (150 rpm)
Drylab Work
Primers
- Margaret, Grace, and Norman
- Worked on redesigning omega cassette primers
- Designed primers to verify rbs and tt
- Reviewed primers, submitted for ordering
Secretion project timeline

- Reviewed timeline with Dr. Presting
- GP suggested that we create a flowchart
