- Sent M213, M218 (dxs+crtEBI) for sequencing. (AM)
- Single (EcoRI) and double (EcoRI+PstI) digests of:
- M198 (SOB2+SU1)
- M201~203 (SOB2-rbs+SU1)
- M204~206 (SOB2+ISO2)
- M207~209 (SOB2-rbs+ISO2). (Yan)
- Single (PstI) digest of P97 (cex mut). (AM)
- M198~M206 run on Gel 80. All failed: single and double digests yielded the same results for all samples. (Yan)
- M207~209 and P97 run on Gel 81. All failed: single and double digests yielded the same results for M207~209. PstI digest of P97 resulted in a smear. (AM)
- Transformation/plating of CHU1727 failed twice. (CKL)
Follow-Up: Sequencing Results
- M212 is pSB1A2-glgC16 with desired prefix and suffix.
- M213 is NOT dxs+crtEBI. Both forward and reverse sequences indicate crtI.
- M218 is NOT dxs+crtEBI. Forward sequence is dxs; reverse sequence is gibberish.
