  • Another miniprep of the S105 gene transformation performed.
  • Optical Density Measurement: 71.6 ng/ul
  • Restriction Digest Verification done of S105 gene with restriction enzymes EcoRI and PstI (Buffer H). Placed in thermocycler for 1 hour at 37 degrees and heat shock 20 minutes at 80 deg C.


  • Performed another ligation of PVJ4 and pSB1AK3. We are still seeing no growth on the PVJ4 or S105 cassette plates.
  • Transformation of the PVJ4 and pSB1AK3 ligation.
