- Another miniprep of the S105 gene transformation performed.
- Optical Density Measurement: 71.6 ng/ul
- Restriction Digest Verification done of S105 gene with restriction enzymes EcoRI and PstI (Buffer H). Placed in thermocycler for 1 hour at 37 degrees and heat shock 20 minutes at 80 deg C.
- Performed another ligation of PVJ4 and pSB1AK3.
- We did see growth for the transformations of pVJ4 and S105 Cassette!! Cultures were placed in the incubator overnight for miniprepping tomorrow.
- Optical Density and conductance test run again with little success. Solution is not clearing up as expected. Cultures will be made from the glycerol stock plates (Colonies 2,3, and 4).
