Another miniprep of the S105 gene transformation performed.
Optical Density Measurement: 71.6 ng/ul
Restriction Digest Verification done of S105 gene with restriction enzymes EcoRI and PstI (Buffer H). Placed in thermocycler for 1 hour at 37 degrees and heat shock 20 minutes at 80 deg C.
Performed another ligation of PVJ4 and pSB1AK3.
We did see growth for the transformations of pVJ4 and S105 Cassette!! Cultures were placed in the incubator overnight for miniprepping tomorrow.
Optical Density and conductance test run again with little success. Solution is not clearing up as expected. Cultures will be made from the glycerol stock plates (Colonies 2,3, and 4).
