Another miniprep of the S105 gene transformation performed.
Optical Density Measurement: 71.6 ng/ul
Restriction Digest Verification done of S105 gene with restriction enzymes EcoRI and PstI (Buffer H). Placed in thermocycler for 1 hour at 37 degrees and heat shock 20 minutes at 80 deg C.
Performed another ligation of PVJ4 and pSB1AK3. We are still seeing no growth on the PVJ4 or S105 cassette plates.
