Things we did today
Wetlab work
Triparental conjugation
- Grace
- Began triparental conjugation between DH5α transformed with pRL1383a, E. coli with RP1, and PCC6803 (see experiment)
- Innoculated BG-11 + 5% LB plates
Purified BBa_B0034 colonies
- Krystle
- Streaked out colonies onto fresh LB+amp100 plates
Preparation for making competent cells
- Krystle & Grace
- Autoclaved detergent-free erlenmeyer flasks
- Innoculated 250 ml SOB w/ DH5α cryostock; shook overnight at 26C with shaking (150 rpm)
Made BG-11+sm+sp plates
- Krystle
- Made 20 plates
- 2.5 μg/ml each of sm and sp, labeled Sm2.5Sp2.5
Made SOB media
- Krystle
- 2 batches (and added antibiotic... oops! will autoclave again to get rid of it)
