Today we started a set of new transformation with various slight differences in order to see if it would help in transformation efficiency.
We tried solving the punches from the registry in either:
- old TE
- fresh TE
- H2O
And with these we either:
- added 4 ul to 50 ul competent cells
- added 50 ul competent cells to all the DNA solution including the punch
As positive controls we transformed with pUC18 in H2O or in old TE with a blanc paper punch added, both incubated with the other samples at 42 for 20'.
