Live stabs
All the live stabs cultures have grown in 5 ml LB + Amp. We've made glycerol stocks, plated them and grown them in 50 ml LB medium + Amp o/n again to perform midiprep tomorrow.
Testing the Primers
The PCR products were run on two different 1.5% agarose gels. Also a marker was run on the gel. The expected band sizes are ~550bp for idi, ~900bp for ispA and ~1200bp for atoB. It is known that bands end up a little higher when running the SybrSafe product in the gel. From these pictures it was concluded that the optimal annealing temperature for IDI and ISPA is 50.5C and 54.5C for ATOB. The pictures are shown below:
