September 5th
3A Assembly
The transformants from the assembly have grown, indicating the assembly has worked. We did a colony PCR on a couple of the transformants, showing insert sizes were ok. (PCR products are ca 315 bp larger then the cut product)
2 colonies which gave large bands will be grown o/n and will be prepped on Monday.
Living stabs
A new living stab has arrived for B0030, which didn't grow earlier. We will grow it o/n as well.
Thermosensitive DNA
A couple of our ordered DNA-parts were delayed, so we requested Geneart to send those finished already. Today we received them and have transformed them on Amp selection.
