A colony PCR on eight colonies for atoB, idi and ispA were performed. The PCR was performed o/n, after the program the machine was programmed to keep a constant temperature of 4 degrees Celsius. The result will be checked on gel tomorrow.

Transformation

The DNA ligation over the weekend has been transformed into TOP10 made chemically competent.

Induction

To test for R0080 function, we're trying to visualize luciferase expression on an SDS-PAGE. Therefore we made a 1.33 M arabinose stock (1000x) for induction [http://tools.invitrogen.com/content/sfs/brochures/710_01619_pBAD_bro.pdf] (page 3) and have grown [http://partsregistry.org/wiki/index.php?title=Part:BBa_K115012 K115012] and [http://partsregistry.org/wiki/index.php?title=Part:BBa_K115011 K115011] o/n both with and without induction.
