Results of the transformation

  • Positive control : pUC19 (1µl; 10pg/ml) --> No transformation
  • Biobricks 2007 : --> succeed of the transformation unlike the delay added in the ice
  • Biobricks 2008 : --> No transformation
          Conclusion : Cells are competents, because there is positive transformation
          Hypothesis : problem with the protocol of DNA extraction from the registery 2008

  • Culture of DH5ɑ in 5ml of LB without Antibiotics, O/N at 37°C

Preparation of medium and solutions

  • 1L of CCMB80
  • 500ml of SOB ( 2 x 250ml)
  • 500ml of SOC (2 x 250ml)
  • 100ml of MgCl2 (1M)
  • 150ml of CaCl2 (1M)
  • 200ml of KOAC (1M)
